The short version

AOD-9604 is a small synthetic peptide copied from the tail end of human growth hormone — specifically residues 177–191, with one stabilizing tweak at the front. The interesting part is what it leaves out: researchers study it because it carries the fat-metabolism side of growth hormone while leaving the growth and blood-sugar effects behind.

What researchers are looking at

Growth hormone does a lot of different jobs at once — it tells the body to grow, it shifts blood sugar, and it nudges fat cells to release stored fat. AOD-9604 is a fragment of that larger hormone, and in lab models it appears to keep only that last job: stimulating fat breakdown (lipolysis) while holding back fat storage.

What draws scientific attention is the separation. Because the fragment lacks the binding regions needed to switch on the full growth-hormone receptor, researchers observe that it does not raise IGF-1 and does not appear to disturb glucose or insulin sensitivity — the two effects that complicate studying whole growth hormone. In the literature, this is framed as isolated lipolytic activity without the broader anabolic signaling.

Most of this evidence comes from animal and test-tube work, where the fragment has been shown to act on fat tissue directly through β3-adrenergic pathways. The findings are best read as what researchers measure in these models rather than proven human outcomes — though AOD-9604 is unusual in this class for having also been carried into human obesity trials.

01Step

Fat release stimulated

In models, the fragment engages β3-adrenergic receptors on fat cells and drives lipolysis — the breakdown of stored fat.

02Step

Fat storage held back

At the same time, lipogenesis — the laying down of new fat — is inhibited within the adipocyte.

03Step

Growth signaling left out

Without the full receptor-binding domains, it raises no IGF-1 and leaves glucose and insulin sensitivity unchanged in studies.

From the lab toward the clinic

AOD-9604 went further than most fragments in this class: it advanced into Phase 2 and Phase 3 human obesity trials, and later received FDA GRAS (Generally Recognized As Safe) designation — placing it well beyond purely preclinical compounds in how far the human data has been carried.
