The short version

Ipamorelin is a tiny five-amino-acid peptide that nudges the pituitary to release the body's own growth hormone. What makes it interesting to researchers is how clean the signal is: in the literature it raises GH while leaving the stress hormones — cortisol, ACTH — and prolactin essentially untouched, which is the opposite of how the earlier growth-hormone peptides behaved.

What researchers are looking at

The body makes its own growth hormone in the pituitary, and it releases it in pulses rather than a steady stream. Ipamorelin is a "secretagogue" — a compound that prompts that release rather than replacing the hormone itself. It works by mimicking ghrelin, the same hunger-and-growth signal the body already uses, and binding to ghrelin's receptor (GHSR-1a) on the hormone-producing cells.

What sets it apart in the research is selectivity. Older growth-hormone peptides tended to spill over and also raise cortisol and prolactin — stress and milk-signaling hormones that complicate any study. Ipamorelin was designed to pull only the growth-hormone lever: in published models it triggers a robust GH pulse with no measurable rise in ACTH, cortisol, or prolactin. Because it works through the body's own pulsing machinery, researchers note it tends to preserve the natural rhythm of release rather than flattening it.

Most of this evidence is preclinical — animal and cell-based studies, including the original work that first characterized it as "the first selective growth hormone secretagogue." The findings are best read as what researchers observe in those models rather than proven human outcomes, but the clean, selective profile is what keeps it a reference point in growth-hormone research.

01Step

Ghrelin's receptor engaged

Ipamorelin mimics ghrelin and binds the GHSR-1a receptor on the pituitary's hormone-producing cells, the somatotropes.

02Step

The release signal fires

Binding triggers an internal calcium-signaling cascade (via phospholipase C and IP3) that tells the cell to release its stored growth hormone.

03Step

A clean GH pulse

Growth hormone is released in a pulse — without the cortisol, ACTH, or prolactin rise seen with earlier secretagogues in the models studied.
